银杏叶黄酮类化合物的测定方法

  银杏(Ginkgo biloba)为我国特有的珍稀植物。银杏叶制剂临床上广泛用于治疗心脑血管疾病[1],目前认为其有效成分为黄酮类(flavonoids)和银杏内酯类化合物,银杏总黄酮的含量可直接反应产品的质量。银杏黄酮按其母核结构可分为3类:(1)5,7,4′-三羟基黄酮醇及其糖苷,重要的化合物为槲皮素(quercetin,Q)、山奈酚(kaempferol,K)、异鼠李黄素(isorhamnetin,I)的没食子酰基单、双、三糖苷(图1);(2)儿茶素类;(3)非糖苷的双黄酮[2](图2和表1)。双黄酮是银杏的特征性成分。
   银杏叶黄酮类化合物的测定最早使用的是分光光度法,通常采用硝酸铝-亚硝酸钠为显色剂,生成铝螯合物在510 nm处测定,以芦丁为对照品[3]。比色法只能粗略评估总黄酮含量,且由于原花色苷元类化合物的干扰,此法的重现性差[4]。高效液相色谱法(HPLC)现已成为测定银杏叶黄酮类化合物的主要方法,毛细管电泳法(CE)也越来越多地应用于黄酮类化合物的测定。另外,还有应用薄层扫描法测定银杏黄酮类化合物的报道[5]。下面就高效液相色谱法和毛细管电泳法做一综述。……
