(上海医科大学药学院药理教研室 200032) 摘要 改良HPLC测定人及大鼠血清中阿昔洛韦浓度的分析方法。使用Waters高效液相色谱仪,μ-Bondapak
C18柱,流动相为甲醇—水(3∶97,V/V),流速1.5mL/min,检测波长254nm,内标物为扑热息痛,血清样品采用甲醇去蛋白。阿昔洛韦血清浓度在0.05~1.6mg/L范围内与峰高呈良好线性关系,大鼠和人血清中相对回收率分别为93.31%及90.69%;最低检测浓度均为0.025mg/L;天内精密度分别为0.83%~2.00%及1.10%~2.58%;天间精密度分别为2.01%~5.90%及3.90%~6.27%。 阿昔洛韦(Acyclovir,简称Acy)为核苷类抗病毒药,它已成为治疗单纯疱疹病毒和带状疱疹病毒感染,特别是重症感染的首选药[1]。本品血药浓度的测定虽已有HPLC方法报道[2~6],但尚不理想,如样品处理过程繁琐,流动相酸度偏高对仪器有损害等。为此,本研究对Acy的HPLC检测条件进行了较为系统的研究,流动相使用普通的甲醇—水体系,试剂价廉低毒,血清样品处理采用直接去蛋白法简便易行。本方法灵敏、精确,可供药代动力学研究及临床血药浓度检测之需。 |

40mg/kg 2h后的血清提取液中,加入一定量的Acy甲醇标准液,测得的色谱峰与没有加入标准液的血清提取液完全重叠,且前者峰高高于后者,证明血清中分离所得化合物为Acy。 表1 Acy在大鼠及人血清中的回收率 |

3.6 天内及天间精密度 制备大鼠及人空白血清的Acy溶液,其浓度分别为0.1、0.4、1.2mg/L血清,各加内标1μg,提取测定,1d内重复测定4次求得天内精密度,每天1次,重复测定4d求得天间精密度,见表2。 表2 Acy在大鼠及人血清中的 |

was set at 254nm.Acetaminophen was used as internal standard.The
serum sample was extracted by a methanol protein precipitation method.Serum
concentration of Acy ranging from 0.05mg/L to 1.6mg/L showed good
linear relationship with the peak ratio and the minimum concentration
detected was 0.025mg/L.The average recoveries in rat and human were
93.31% and 90.69% respectively.The intraday precision was <3% and
interday precision was <7%. |
