摘要 对生长激素的检定和质量控制的重要指标之一是效价测定,其传统经典的方法为在体生物测定法—体重法和胫骨法。随着基因工程技术的发展和质量标准的不断完善,对rhGH的效价测定已用理化分析方法代替生物测定方法。本实验通过对进口和国产的多批rhGH制剂和原料的效价的对比实验,证明生物测定和理化测定的结果基本吻合,具有良好的相关性,为理化测定最终代替生物测定提供了实验依据。 人生长激素(hGH)过去由人脑垂体提取,其成分不单一[1],对其检定只能依赖生物测定─去垂体大鼠体重法[2~4]和去垂体大鼠胫骨法[5]。随着科学的进步,DNA重组生长激素(rhGH)产品日趋成熟,纯度已达95%以上,生产工艺和产品已趋稳定,人们不断研究用简单、快速、灵敏、精确的理化方法代替费时、误差大、损害动物的生物测定[6,7]。现已载入BP(1993)[8]和EP(1997)[9],并都明确规定必须用适当有效的增进生物生长的方法,来证明生产制造过程中的rhGH产品不得少于2.5IU/mg的生物活性,并由国家卫生行政机构批准执行。在此前提下常规生产中,rhGH的效价测定可用理化方法(HPLC)代替。本文通过对进口和国内生产的多批rhGH制剂和原料的效价进行了生物测定和理化测定,结果表明两法基本吻合,具有良好的相关性,为理化测定最终代替生物测定法提供了实验依据。 表1 生物测定和理化测定效价的比较* |

Zhaopeng and Xu Kangsen Abstract:The potency for growth hormone(GH)
is one of the important parameter in quality control.Historically,it
depended on the use of in vivo bioassay.Today,it has been suggested
that the in vivo bioassay of GH could be replaced with physico-chemical
assay in routine control.Size-exclusion HPLC method was compared
with GH bioassay in our laboratory.The potency of various rhGH preparations
and materials from many manufacturers at home and abroad was determined
by in vivo bioassay including weight gain and tibial width in hypophysectomized
rat and size-exclusion HPLC to examine the relative correlation
of the two methods.Experimantal results showed that the contents
obtained by HPLC method is generally in agreement with the potency
by bioassay.Based on the study,the bioassay of rhGH could be replaced
with HPLC in routine control. |
