摘 要 DETERMINATION OF LOFLOXACIN BY HPLC IN HUMAN SERUM AND STUDY OF PHARMACOKINETICS Jiang Lifu,Zheng Yi,Ma Yaping,Xiao Yong Abstract AIM A sensitive HPLC method has been
developed for the determination of Lofloxacin in human serum.METHODS Lofloxacin analysis was carried
out by using ODS column (150x4.6mm,ID).The mobile phase consisted
of 0.03mol.L-1
phosphoric acid:acetonitrile:tetrabuty lammon hydroxide (80∶7∶1,v/v).Pipemidic
acid was used as an internal standard.RESULTS The method has good linearity betwcen 0.4~4.0μg.ml-1.The regression equation is Y=2.330X-0.100,r=0.990
4.Recovery rate varied from 91.7% to 102.0%.CONCLUSION The method can be used in pharmcokinetics. 芦氟沙星为一种新型广谱氟喹诺酮类国家级二类抗菌药。目前不少医院已开始应用于临床,但有关该药的研究多为临床疗效观察。笔者参考了国外有关文献,建立了快速,简便易行的血清芦氟沙星浓度的HPLC测定法,适用于芦氟沙星的临床药动学、药效学及药物相互作用方面的研究。 1 仪器与试药 2 方法与结果 图1 健康志愿者口服芦氟沙星后药-时曲线 将药-时数据用PKBP-N1药代动力学程序在IBM微机处理,曲线拟合后选择房室模型,药动学参数计算结果见表1。 表1 口服芦氟沙星人体内药代动力学参数 |
