摘要 建立一种HPLC法快速测定头孢拉定血清浓度。取全血1 mL,离心,20 μL直接进样。色谱柱:Hi-Pore Reversed Phase
Column(4.6 mm×250 mm),预柱:Bio-sil ODS-5s Cartridges,分析流动相10 %乙腈,预流动相0.9
%氯化钠,切换时间2 min,检测波长254 nm。药物保留时间7.05 min,分离良好;血清浓度2~100 μg/mL范围内线性良好,r=0.9997;回收率、日内差、日间差均较好。该法快速、准确、可行。 头孢拉定(Cefradine,CEF)是一种可供口服、肌注、静注的头孢菌素,其对β-内酰胺酶稳定,肾毒性低,被广泛用于临床。鉴于柱切换高效液相色谱法[1]的优点,本文建立了头孢拉定柱切换高效液相色谱法,样品不经处理,可直接进样,具有快速、简捷、方便等优点,尤适合于头孢拉定的动力学测定及血药浓度监测。 |

50 mg,制成浓度为20、50、100、250、500、1000 μg/mL系列,精取上述标准液各50

μg/mL血清样品,放于冰箱中冷藏,分别于早晨九点、中午一点、晚上六点取样测定,得日内差3.09
%(n=3),连续3日取样测定,得日间差3.16 %(n=3)。 |

mm×250 mm)was used as analytical column.The mobile phase consisted
of acetonitrile-water(1∶9).The eluent was detected at 254 nm.Linear
range was 2~100 μg/mL.The recovery rate was 98.4 %~99.34 %.The
RSD within a day was 3.09 % and between days was 3.16 %.The method
was found to be simple,quick and accurate for determination of cefradine
in human serum. |
