SLE患者HSP90基因表达及其与IL-6相关性的研究

摘 要:

目的:通过检测系统性红斑狼疮(SLE)患者外周血中热休克蛋白90(HSP90)基因mRNA及IL-6的水平,探讨SLE患者HSP90基因的表达及其与IL-6间存在的相关性。方法:分别使用原位杂交法、双抗体夹心ELISA法检测45例SLE患者外周血中HSP90 mRNA和IL-6的表达,并进行直线相关性分析。取15份健康人血样进行正常对照。结果:SLE活动期、非活动期PBMC中HSP90 mRNA表达的平均光度值分别为0.350±0.225,0.221±0.040,均显著高于正常对照组0.114±0.009(P〈0.01,P〈0.c15)。SLE活动期HSP90 mRNA表达显著高于SLE非活动期(P〈0.05)。SLE活动期、非活动期血清IL-6水平分别为(16.82±1.79)pg/mL,(12.58±1.12)pg/mL,均显著高于正常对照组(10.13±2.95)pg/mL(P〈0.01,P〈0.05)。SLE活动期IL-6水平显著高于SLE非活动期(P〈0.01)。IL-6的表达与HSP90 mRNA的含量之间呈直线正相关关系(r=0.413,P〈0.01)。结论:SLE患者体内存在IL-6的过度表达,可导致HSP90 mRNA表达的增多,进而导致HSP90蛋白增高,高表达的HSP90可参与SLE的自身免疫反应,这可能在SLE的发病机制中起着一定的作用。

Study on the correlation of HSP90 gene and its correlation to IL- 6 in patients with systemic lupus erythematosus

Abstract:

Objective: To study the expression of HSP90 gene and its correlation to IL- 6 in patients with systemic lupus erythematosus (SLE) through detecting the expression of HSP90 gene in the peripheral blood mononuclear cells (PBMC) and IL-6 in the serum from SLE patients. Methods: Applying in situ hybridization technique to detect the expression of HSP90 mRNA in the PBMC from 45 SLE patients and 15 healthy controls, and IL - 6 expression in the serum in the same groups was measured by ELISA. Results: Semi - quantitative measurement of HSP90 mRNA expression in the blood specimen showed that the mean luminosity in active SLE patients, inactive patients and healthy people was 0.350 + 0.225, 0.221 + 0.040 and 0.114 + 0.009 respectively. Active SLE patients and inactive SLE patients had higher levels of HSP90 mRNA expression in PBMC compared with healthy controls( P 〈 0.01, P 〈 0.05), and active SLE patients had higher levels of HSP90 mRNA expression compared with inactive SLE patients ( P 〈 0.05). The levels of IL- 6 in sera of active SLE patients, inactive patients and healthy people was 16.82 + 1.79 pg/ mL, 12.58 + 1.12pg/mL and 10.13 + 2.95 pg/mL respectively. Active SLE patients and inactive SLE patients had hiller levels of IL - 6 in sera compared with healthy controls( P 〈 0.01, P 〈 0.05), and active SLE patients had higher levels of IL- 6 compared with inactive SLE patients( P 〈 0.01). A linear positive relationship was found among expression of IL- 6 and levels of HSP90 mRNA( r = 0.413, P 〈 0.01). Conclusion: There is overexpression of IL- 6 in SLE patients, elevated levels of IL- 6 in SLE patients can induce the expression of HsP90mRNA increase, and in turn results in elevated levels of HSP90 protein, overexpressional HSP90 participates in the autoimmune reaction of SLE, which may play a certain role in the pathogenesis of SLE.
