  摘 要:目的 构建表达乙型肝炎病毒(HBV)S蛋白的重组质粒pCR3.1-S作为核酸疫苗,观察重组人白细胞介素2(rhIL-2)作为佐剂对其诱导BALB/c小鼠产生免疫应答的影响。方法 分别以ELISA法、3H-TdR掺入法、Cr51 4h释放法等测定免疫小鼠的血清抗HBs、淋巴细胞增殖活性、淋巴细胞杀伤效应,初步研究不同免疫组体液和细胞免疫应答的差异。结果 血清抗HBsELISA效价,rhIL-2组与对照组相比较差异不显著(P>0.05);淋巴细胞增殖指数、淋巴细胞杀伤率,1000U(只次)rhIL-2/组和2000U(只次)rhIL-2/组与对照组相比较差异显著(P<0.05),而500U(只次)rhIL-2/组与对照组相比较无显著差异(P>0.05)。结论 一定剂量的rhIL-2作为佐剂可增强HBV核酸疫苗的免疫效应。

Effects of different doses of interleukin 2 as adjuvants on immune responses induced by nucleic acid vaccines of hepatitis B virus

  Abstract:Objective To construct recombinant plasmids of pCR3.1-S encoding protein S of hepatitis B virus as nucleic acid vaccines and investigate the effects of recombinant human interleukin 2 as adjuvants on immune responses induced by HBV nucleic acid vaccines in BALB/c mice. Methods  Anti-HBs antibodies to HBV major protein was measured by ELISA assay, and the activities of lymphocyte proliferation and cytotoxity were detected by  3H-TdR incorporation assay and 4-hour 51Cr release assay. Humoral and cellular immune responses of different groups were then compared. Results  The titers of anti-HBs antibodies in serum of rhIL-2 group mice were not significantly higher than those of control group mice (P>0.05). The difference of the activities of lymphocyte proliferation and cytotoxity between of control group mice and rhIL-2 group mice (every mouse with 500 U one time) was not significant (P>0.05). However, stimulation index and specific lysis rate of lymphocyte of rhIL-2 group mice (every mouse with 1 000 U or 2 000 U one time) showed significant difference compared with htose of control group mice (P<0.05). Conclusion  Recombinant plasmids of pCR3.1-S as HBV nucleic acid vaccines could induced strong humoral and cellular immune responses in BALB/c mice. rhIL-2 acts as adjuvants to increase the potency of HBV nucleic acid vaccine in some certainty.
