摘 要 用RP-HPLC以安宫黄体酮为内标物同时测定板蓝根、大青叶中靛蓝、靛玉红的含量.色谱条件:色谱柱为Spherisorb C18柱;流动相为甲醇-水(62∶38,V∶V)检测波长280 nm;靛蓝和靛玉红的平均回收率分别为(99.0±1.1)%和(104.2±3.5)%.本法操作简便,结果准确. Determination of the Content of Indigo and Indirubin in the Indigowoad Root and Leaves by RP-HPLC Sun Lixin,Tang Hong,Yin Ping,Ning Lili,Bi Kaishun Abstract An analytical method for determination of indigo and indirubin in the indigowoad root and leaves by RP-HPLC was developed.The HPLC conditions were as follows:spherisorb C18 column(4.6 mm×250 mm)the mobile phase,methanol-H2O(62∶38,V∶V)detection wavelength,280 nm;internal standard,medroxyprogesterone acetate.The mean recoveries for the indigo and indirubin were(99.0±1.1)% and(104.2±3.5)%,respectively.The method was simple and accurate. 板蓝根为十字花科植物菘蓝(Isatis indigotica Fort.)和爵床科植物马蓝Baphicacanthus cusia(Nees)Bremek.的干燥根〔1〕.大青叶为菘蓝、马蓝和蓼科草本植物蓼蓝(Polygonum tinctorium Ait.)的枝和叶〔2〕.板蓝根和大青叶均具有清热解毒、抗菌消炎、凉血消斑等功效,二者在中药成方中经常配伍入药,如金青感冒颗粒,小儿退热颗粒,复方板蓝根颗粒等.板蓝根和大青叶的有效成份都是靛蓝(indigo)和靛玉红(indirubin)〔3〕.靛玉红对治疗慢性粒细胞白血病有较好疗效.靛蓝、靛玉红的含量测定方法已有氧化滴定法、直接比色测定法、薄层色谱法、双波长分光光度法、反相高效液相色谱法〔4〕.其中反相高效液相色谱法采用外标法定量,对获得操作的重复性来说有其不足之处,本文则采用安宫黄体酮为内标物以内标法定量,结果令人满意. 1 仪器与材料 日本岛津公司高效液相色谱仪LC-6A系列;LC-6A溶剂输送泵;SPD-6AV紫外-可见分光检测器;LC-R3A数据处理装置;7125 20μL定量进样阀. 2 方法与结果 2.1 分离条件 Tab.1 Content of indigo and indirubin in the idigowoad root and leaf from various origins |

作者试用联苯、萘、苯甲酸苯酯、安宫黄体酮等作为内标物,从分离度和保留时间上看,以安宫黄体酮为宜;避光条件下,24 h内板蓝根、大青叶中靛蓝、靛玉红在氯仿中稳定,时间过长,靛蓝、靛玉红分解,故应保证在24 h内完成实验;作者首次采用内标法,准确地测定了板蓝根、大青叶中靛蓝、靛玉红的含量,结果准确、可靠,重复性好.
