(浙江省丽水市中心医院 丽水 323000) 摘要:本文采用反相高效液相色谱法测定人血浆中格列齐特浓度。固定相:ODS-C18;流动相∶乙腈-甲醇-水(35∶20∶45)的混合溶液,并用磷酸调节pH至3.5;流速:1.0
ml.min-1;紫外检测波长:228 nm;方法回收率大于95%,日内、日间相对标准偏差小于8%。血浆的最低检测限为0.05
μg.ml-1;线性范围:0.5~12
μg.ml-1,r=0.9995。 Determination of Gliclazide in Human
Plasma by RP-HPLC ABSTRACT:A reverse phase RP-HPLC method has
been successfully used to determine Gliclazide in plasma.Ananalytical
column was packed with ODS-C18.The mobile phase was
acetonitrile—methanol—water(35∶20∶45)(and phosphate is used to adjust
pH=3.5,flow rate of 1.0 ml.min-1 UV detection was performed at
228nm.The average recovery was >95%.The respense value was linear between
0.5 μg.ml-1~12 μg.ml-1 and r was

2 结果 2.1 线性关系与灵敏度 取7支试管分别加空白血浆1.0 ml,1支试管作空白对照,另6支分别另入GL储备液使含GL为0.5、1.0、2.0、5.0、10.0、12.0 μg.ml-1的浓度系列,按前法提取进样。以GL峰面积x为纵座标,相应浓度Y为横座标,作标准曲线(见图2)。其直线回归方程为:Y=0.02 32x-0.04681(r=0.9995)。 |

2.3 精密度 取3支试管,按血样品处理法分别配成浓度为0.5、5、12
