(军事医学科学院毒物药物研究所 北京 100850) 摘要 建立了比格犬血浆样品中托烷司琼的反相高效液相色谱检测法。血浆样品,加入盐酸三环哌酯作内标,用乙酸乙酯提取,经稀盐酸反洗,再用二氯甲烷提取。色谱条件:C8柱;流动相:甲醇-水(55∶45),加0.3
%三乙胺,用冰醋酸调pH 7.2;检测波长283 nm。线性范围5.45~700 ng/mL, 最低检测浓度为3.0
ng/mL ,回收率为79.9 %~85.8 %,日内RSD为1.06 %~4.27 %,日间RSD为2.94
%~9.89 %。采用本法对比格犬口服托烷司琼的药代动力学进行了研究。 |

μL,分别用犬空白血浆稀释至1.0 mL,使终浓度分别为10.9、175和700 ng/mL,按提取方法提取,进样测定托烷司琼的峰面积,与相同浓度托烷司琼的对照品溶液直接进样测得的峰面积比较,计算方法的提取回收率,结果3种浓度的回收率分别为85.85
%,79.90 %和83.63 %,RSD分别为7.66 %,8.74 %和4.72 %,n=4。 |

liquid chromatographic method is described for the determination
of tropisetron in plasma of beagle dogs.Tropisetron in plasma was
extracted with ethyl acetate after the addition of the internal
standard tricyclopinate hydrochloride.The organic phase was re-extracted
with dilute hydrochloric acid and the aqueous phase was back-extracted
with dichloromethane.After the extract was evaporated to dryness,the
residue was dissolved in methanol and then separated on a C8 column.The mobile phase consisted of methanol-water(55∶45)with
0.3 % triethylamine and was adjusted to pH 7.2 by acetic acid.The
detector was set at UV 283 nm.The linear range of the calibration
curve was 5.45~700 ng/mL and the minimal detectable concentration was
3.0 ng/mL.The recovery of the method was 79.9 %~85.8 %.The within-day and between-day
precisions were 1.06 %~4.27 % and 2.94 %~9.89 %,respectively.The method has been applied to the
pharmacokinetic study of tropisetron in beagle dogs after po 20
