HPLC分离后,与其它技术联用,如HPLC—MS(质谱)[12],HPLC—IR(红外光谱)[13],HPLC—FD—MS(场解吸质谱)[15],HPLC—MIR—IR(多内反射红外光谱)[15]等,可以更好地对色谱峰进行定性,保证人参单体皂苷分析的准确性。 3 样品处理
人参的样品处理一般以醇(甲醇或乙醇)提取,以醚或氯仿脱脂后,以Sep-Pak
C18柱或大孔吸附树脂处理,或以水饱和正丁醇多次萃取净化,减压蒸干,以流动相或甲醇等溶剂定容后进行色谱分析。 4 结语
综上所述,高效液相色谱法(HPLC)分析人参皂苷,在分离、检测、样品处理等方面,目前已取得一定进展,但仍不能满足实际分析的需要,如分离效果、分析的灵敏度、精确度等方面均需进一步提高,尤其是现有的HPLC分析方法检测限为ng级,还不能满足今后的分析需要,因此,如何提高灵敏度,研究新的HPLC检测方法,是今后工作的方向之一,其中寻找灵敏的衍生试剂,开发柱前、柱后衍生反应,使紫外吸收灵敏度提高,使用电化学检测或荧光检测以及开发新的高灵敏检测器都有可能进一步提高灵敏度。 参考文献 1 Petersen T G,et al.Utilizing column selectivity
