Bondapak C18色谱柱,甲醇—0.025
mol/L磷酸二氢钾(42∶58,pH 2.5)为流动相,295 nm为检测波长,非那西丁为内标,样品用甲醇沉淀蛋白,经0.45
μm滤膜过滤后富集进样。该法灵敏、简便、准确,氧氟沙星在0.25~10
μg/mL浓度范围内线性关系良好(r=0.9997),检测限为0.15 μg/mL(S/N=3∶1),平均回收率为101.1
%,不同浓度水平的日内和日间精密度测定结果RSD均小于10 %。采用该法对8名健康受试者口服氧氟沙星片后的药代动力学和生物利用度进行了研究。 氧氟沙星(氟嗪酸,Ofloxacin,OFLX)是目前广泛应用于临床的第三代喹诺酮类药物,主要用于治疗呼吸道、泌尿道、肠道、皮肤、关节及软组织等多种系统的感染,具有抗菌谱广、作用强,与多种抗菌药物间无交叉耐药性的特点。对OFLX在人体内血药浓度的测定,目前国内外文献报道一般采用HPLC—荧光检测法或生物分析法[1~3]。本文参考有关文献[4,5]并加以改进,建立了灵敏度较高、线性良好、操作简便的HPLC—紫外检测法,并以康利必妥片为参比制剂,对氧氟沙星片在正常人体内的药代动力学及生物利用度进行了研究。 |

method for the determination of ofloxacin(OFLX)in human plasma.Using
a μ-Bondapak C18
column(25 cm×4.6 mm,10 μm)under room temp,the mobile phase consisted
of methanol—0.025 mol/L KH2PO4(42∶58,pH
2.5)and the flow rate was 1.0 mL/min.The UV detection wavelength
was 295 nm.Using phenacetin as internal standard and deproteined
by adding 1.5 mL methanol.The precipitated mixture was shaken and
then centrifuged at 3000 r/min for 10 min,after filtration by 0.45
μm filter membrane,the filtration was evaporated to dryness.The
residue was dissolved in 0.1 mL methanol and 20 μL of the soln.was
injected into the system.Within 0.25~10 μg/mL,OFLX had a good
linearity(r=0.9997).The detection limit was 0.15 μg/mL(S/N=3∶1)and
the mean recovery of OFLX was 101.1 %.The within-day and day-to-day
precision expressed by RSD was less than 10 % at three drug levels. |
