摘要 化学法提取的蚕蛹油经甲酯化后,采用气相色谱/质谱(GC/MS)技术,对蚕蛹油中脂肪酸的组分进行了分析,以十九烷酸(19∶0)甲酯作内标物,气相色谱法(GC)对蚕蛹油中α-亚麻酸进行了定量测定。结果表明蚕蛹油中α-亚麻酸的含量高达72.80 %。本研究为α-亚麻酸的定量测定提供了实验依据。 蚕蛹为蚕蛾科昆虫家蚕蛾Bombyx mori的蛹,具有很高的营养价值[1,2]。在医学上蚕蛹具有降低血胆固醇,治疗冠状动脉供血不足等效果[3]。本文采用GC/MS法对蚕蛹油中的脂肪酸的组分进行了分析和鉴定,发现蚕蛹油中主要成分为α-亚麻酸(α-Linolenic acid,α-18∶3), 并用气相色谱内标法对 其α-亚麻酸的含量进行了测定。 |

图2 蚕蛹油脂肪酸和内标气相色谱图 3.2.2 标准曲线制作 各取不同浓度梯度的α-亚麻酸标准液(0.25、0.5、1、2、4 μg/μL)与内标液(2 μg/μL)等比例混合,各取混合液1 μL,按GC条件进样测定。绘制标准曲线,在0.125~2.000 μg/μL范围内,呈良好线性。回归方程: (本文于1997年9月22日收到) Content Determination of α-Linolenic Acid in Can Yong Oil Lu Ping,Lai Bingsen,Wang Yingqiang and Tan Yafang Abstract:Can Yong oil was extracted by chemical method.The fatty acid(FA)in Can Yong oil was saponificated by KOH-MeOH and was modified to methyl esters by means of BF3-MeOH,and was checked with thin-layer chromatography.The compositions of fatty acids were analyzed by GC/MS.The method of GC internal quantification of α-Linolenic in Can Yong was established,using internal standard of C19∶0.A linear relationship was obtained in the concentration rage of 0.125~2.000 μg/μL and the correlation coefficient was 0.9830.The content of α-Linolenic acid was 72.80 %.The study may give a test method for determination of α-Linolenic. |
