摘 要:用高效液相色谱法测定血浆中鹿蹄草素的浓度。方法:采用YWG-C18色谱分析柱(4.6mm×250mm,5μm),WatersTM486可变波长紫外检测器,甲醇-水(40∶60)为流动相,香草醛为内标。血液样品用乙醚提取纯化后进样,在220nm波长处检测。结果:鹿蹄草素线性范围为1.0~100.0mg.L-1(r=0.9953),最低检测量0.1μg,最低检出浓度5.0mg.L-1,平均方法回收率98.36%,精密度考察鹿蹄草素日内、日间RSD均小于4%。结论:本方法专属性高,快速,准确,重复性好。

AbstractObjectiveA HPLC method is described for the analysis of pyroline in pig plasma.Methods:The YWG-C18 column (4.6 mm×250 mm,5 μm) was used,The mobile phase consisted of methanol-water (4060),at a flow rate of 0.7 mL.min-1.Vanillin was adopted as the internal standard at UV absorption of 220 nm.Results:The ranges of pyroline calibration curves were 1.0100.0 mg.L-1(r=0.995 3). Detection limits were 0.1 μg and the lowest detectable concentration of pyroline was 5.0 mg.L-1,while the average recovery of the method was 98.36%.The RSD of intra-day and inter-day assays was all less than 4%.Conclusion:The method is quick,precise and good repeat.
