摘 要:本文报道了胶束电动毛细管色谱分离、汞灯诱导荧光电荷耦合器件检测分析氨基酸和磷酸化氨基酸的4-氟-7-硝基苯-2-?-1,3-丫唑衍生物。研究表明,在pH9.35的10mmol/L硼砂和10mmol/L十二烷基硫酸钠的电泳缓冲介质中,5种氨基酸和3种磷酸化氨基酸在10min内完全分离,检测灵敏度为1.21×10-8~5.21×10-8mol/L,分离效率达7.3×105~3.0×105/m理论塔板数,结果令人满意。

Separation of Amino Acids and Phosphoamino Acids as Their Derivatives by Micellar Electrokinetic Capillary Chromatography

AbstractThe separation of amino acids and phosphoamino acids as their 4-fluoro-7-nitroben-2-oxa-1,3-diazple fluorescent derivatives was achieved by micellar electrokinetic capillary chromatography with lamp induced fluorescence-cooled charged coupled device detection. Some parameters such as precapillary derivatization conditions and separation conditions were discussed and optimized. With a buffer of pH 9.35 10 mmol/L borate containing 10 mmol/L SDS, the 5 amino acids and 3 phosphoamino acids were separated in 10 min. The detection limits were 1.21×10-85.21×10-8 mol/L with separation efficiency of 7.3×1053.0×105/m.
