摘 要:目的:建立反相高效液相色谱法测定人血清中5-氟尿嘧啶浓度。方法:血清样品用乙酸乙酯提取,水浴氮气吹干,残留物用水溶解后进样。色谱柱为C18柱(250mm×4.6mm),水为流动相,流速1.0mL·min-1,紫外检测波长273nm,桂皮酸为内标。结果:本法最低检测浓度为0.05μg·mL-1,线性范围为1.0~50.0μg·mL-1,日内RSD为2.7%~4.1%,日间RSD为3.8%~4.7(n=4)。结论:该法适用于5-氟尿嘧啶的药代动力学研究及临床血药浓度检测。试验结果表明,该方法经济、简便、快速、灵敏度高、重现性好。
A Simple and Rapid HPLC Determination of 5-Fluorouracil in Human Serum LIU Chuan-hua(Laboratory
of Molecular Biology of Medical College,Shandong University,Jinan 250012) Abstract:Objective:To develop a simple and rapid HPLC method for the determination of 5-fluorouracil (5-Fu)concentration in human serum.Methods:The serum samples were extracted with ethyl acetate.The solvent was evaporated to dryness under a gentle stream of N2 at 40 ℃ in a water bath.The analysis involved a C18 column as stationary phase and distilled water as mobile phase.The flow rate was 1.0 mL·min-1,UV detection wavelength was at 273 nm.Cinnamic acid was used as the internal standard.Results:The minimal detectable serum drug concentration was 0.05 μg·mL-1.The calibration curve was linear over a range from 1.0 μg·mL-1 to 50.0 μg·mL-1 with a correlation coefficient of 0.999 4.Intra-day and inter-day RSD were 2.7%~4.1% and 3.8%~4.7%(n=4) respectively.Conclusion:The method is economic ,simple,rapid,sensitive and accurate.
